RESUMO

Temperature regulates nonradiative processes in luminescent materials, fundamental to luminescence nanothermometry. However, elevated temperatures often suppress the radiative process, limiting the sensitivity of thermometers. Here, we introduce an approach to populating the excited state of lanthanides at elevated temperatures, resulting in a sizable lifetime lengthening and intensity increase of the near-infrared (NIR)-II emission. The key is to create a five-energy-level system and use a pair of lanthanides to leverage the cross-relaxation process. We observed the lifetime of NIR-II emission of Er3+ has been remarkably increased from 3.85 to 7.54 ms by codoping only 0.5 mol % Ce3+ at 20 °C and further increased to 7.80 ms when increasing the temperature to 40 °C. Moreover, this concept is universal across four ion pairs and remains stable within aqueous nanoparticles. Our findings emphasize the need to design energy transfer systems that overcome the constraint of thermal quenching, enabling efficient imaging and sensing.

RESUMO

Different data types often occur in psychological and educational measurement such as computer-based assessments that record performance and process data (e.g., response times and the number of actions). Modelling such data requires specific models for each data type and accommodating complex dependencies between multiple variables. Generalized linear latent variable models are suitable for modelling mixed data simultaneously, but estimation can be computationally demanding. A fast solution is to use Laplace approximations, but existing implementations of joint modelling of mixed data types are limited to ordinal and continuous data. To address this limitation, we derive an efficient estimation method that uses first- or second-order Laplace approximations to simultaneously model ordinal data, continuous data, and count data. We illustrate the approach with an example and conduct simulations to evaluate the performance of the method in terms of estimation efficiency, convergence, and parameter recovery. The results suggest that the second-order Laplace approximation achieves a higher convergence rate and produces accurate yet fast parameter estimates compared to the first-order Laplace approximation, while the time cost increases with higher model complexity. Additionally, models that consider the dependence of variables from the same stimulus fit the empirical data substantially better than models that disregarded the dependence.

RESUMO

Our previous studies showed that S100A9 was overexpressed in glioma and promoted tumor growth. However, S100A9 can also be secreted by tumor cells to regulate the tumor microenvironment (TME). In this study, we aimed to explore the functions of glioma derived-S100A9 in microglial M2 polarization, resulting in inhibition of CD8+ T lymphocytes and promotion of immunosuppression. We first showed that glioma exhibited higher expression and secretion of S100A9 than astrocytes. After knocking down S100A9 in two glioma cell lines, the secretion of S100A9 was repressed. Then, the medium was collected and considered as conditioned medium (CM), which was incubated with microglia. We found that glioma-derived S100A9 drove microglial M2 polarization and increased TGFß1 secretion. These molecular mechanisms were related to the interaction of S100A9 with αvß3 integrin and the subsequent activation of AKT1 in microglia. Furthermore, we demonstrated that S100A9-induced M2 microglia negatively affected cell viability, IL-2 and IFN-γ secretion, together with increased early apoptosis in CD8+T lymphocytes via TGFß1. Additionally, glioma cells were implanted into mouse brains, and we confirmed that S100A9 stimulated microglial M2 polarization, enhanced TGFß1 levels and repressed CD8+ T lymphocytes in orthotopically transplanted tumors. In human glioma samples, S100A9 expression was positively associated with CD206 expression, but negatively correlated with CD8+T lymphocyte accumulation in the TME. Our data indicated that glioma-derived S100A9 has a promising ability to manipulate non-malignant cells and promote immune evasion in the TME, providing valuable insight into the mechanism by which S100A9 participates in the progression of glioma.

RESUMO

Problem-solving is a critical aspect of intelligence that has become increasingly important in modern society. Mapping out the determinants of success in problem-solving helps understand the underlying cognitive processes involved. This article focuses on two key cognitive processes in problem-solving: non-targeted exploration and planning. We generalize previously defined indicators of planning and non-targeted exploration across tasks in the 2012 Programme for the International Assessment of Adult Competencies and examine the internal construct validity of the indicators using confirmatory factor analysis. We also investigate the relationships between problem-solving competency, planning, and non-targeted exploration, along with the specific dependence between indicators from the same task. The results suggest that (a) the planning indicator across tasks provides evidence of internal construct validity; (b) the non-targeted exploration indicator provides weaker evidence of internal construct validity; (c) overall, non-targeted exploration is strongly related to problem-solving competency, whereas planning and problem-solving competencies are weakly negatively related; and (d) such relationships vary substantially across tasks, emphasizing the importance of accounting for the dependency of measures from the same task. Our findings deepen our understanding of problem-solving processes and can support the use of digital tools in educational practice and validate task design by comparing the task-specific relationships with the desired design.

RESUMO

Flooding is one of the most widespread and catastrophic natural disasters. The exposure of cropland to floods is directly related to the quality of cropland and food security, so it is particularly important to map the spatiotemporal evolution of this exposure, with a specific focus on longer time series and higher resolution scales. This study is the first of its kind to analyse the worldwide spatiotemporal variability of Cropland Exposure to Flooding (CEF) with the 30 m resolution of Global Land Analysis & Discovery (GLAD) dataset during 2000-2019. The findings indicate that: (1) the global CEF area increased by a total of 83,429.50 km2 or 7.75 %, from 2000 to 2019; (2) only North America's CEF showed a downward trend, and the region with the largest increase in CEF was South Asia; (3) the CEF in 23 river basins, including Ganges, Indus, Mississippi, Yangtze, and Danube, accounted for 79.88 % of the global total in 2019P; (4) in 2019P, China had the largest CEF globally, reaching 239,525.07 km2. The fastest growing CEF was India, contributing 16.36 % of the global CEF growth. The CEF of United States experienced a reduction trend; (5) two constructed indicators were used in evaluating the CEF of countries worldwide, and a total of 46 countries are considered to be at the highest level of risk, mainly in Europe and Asia. Based on these conclusions, we carried out a cold/hot spot analysis to reveal the spatial heterogeneity and possible driving factors in this phenomenon, and we offer management suggestions to limit the risks to cropland in the floodplains.

RESUMO

Cerebral edema (CE) exerts an important effect on brain injury after traumatic brain injury (TBI). Upregulation of transient receptor potential melastatin 4 (TRPM4) in vascular endothelial cells (ECs) results in damage to capillaries and the blood-brain barrier (BBB), which is critical for the development of CE. Many studies have shown that 9-phenanthrol (9-PH) effectively inhibits TRPM4. The current study aimed to investigate the effect of 9-PH on reducing CE after TBI. In this experiment, we observed that 9-PH markedly reduced brain water content, BBB disruption, proliferation of microglia and astrocytes, neutrophil infiltration, neuronal apoptosis and neurobehavioral deficits. At the molecular level, 9-PH significantly inhibited the protein expression of TRPM4 and MMP-9, alleviated the expression of apoptosis-related molecules and inflammatory cytokines, such as Bax, TNF-α and IL-6, near injured tissue, and diminished serum SUR1 and TRPM4 levels. Mechanistically, treatment with 9-PH inhibited activation of the PI3K/AKT/NF-kB signaling pathway, which was reported to be involved in the expression of MMP-9. Taken together, the results of this study indicate that 9-PH effectively reduces CE and alleviates secondary brain injury partly through the following possible mechanisms: ①9-PH inhibits TRPM4-mediated Na + influx and reduces cytotoxic CE; ②9-PH hinders the expression and activity of MMP-9 by inhibiting the TRPM4 channel and decreases disruption of the BBB, thereby preventing vasogenic cerebral edema. ③9-PH reduces further inflammatory and apoptotic damage to tissues.

RESUMO

The abandonment phenomenon is familiar in China. It threatens food security and seriously affects the sustainable development of society, the economy, and the natural environment. However, monitoring and mapping abandoned cropland on a large scale remains a challenge because of its complex land-use change process. According to the World Food and Agriculture Organization, cropland not used for agricultural production for more than 5 years is defined as abandoned cropland. This study uses the Landsat high-precision long-time series product to detect cropland nationwide from 1990 to 2019 by using the sliding window method on Google Earth Engine to profile the spatial distribution, intensity, trend, frequency, and recultivation of abandoned cropland. Between 1992 and 2015, the results illustrate that the total area of abandoned cropland in China was 559,170.26 km2, 18.59 % of the cropland area. Excluding the recultivated cropland area, China's abandoned cropland area amounted to 392,156.24 km2, 13.03 % of the total cropland. Cropland abandonment is mainly distributed in the second terrain gradient in midwestern and southwestern regions such as Inner Mongolia and Gansu. It rarely happens in western and eastern coastal areas. A high abandonment rate area usually has high elevation and slope. The light index is negatively correlated with the abandonment rate in suburban areas. This study is the first to map the spatiotemporal distribution of abandoned cropland in China with high precision at 30 m resolution. It provides an important basis for policies regarding the recultivation of abandoned cropland.

RESUMO

BACKGROUND: Improper use of strychnine can cause death. The aim of this study was to identify and evaluate toxic mechanisms of action associated with active compounds in strychnine using a network toxicology approach, and explore potential pathogenic targets. METHODS: In the present study, strychnine target and central nervous system-related gene set were established using the Traditional Chinese Medicine Systems Pharmacology (TCMSP) database and four disease gene databases (Genecards, OMIM, PharmGkb, TTD). An "ingredient-target" interactive active network map was constructed using Cytoscape software (version 3.8.0). Functional enrichment analysis was performed based on the hub genes. A protein-protein interaction network was constructed using STRING database. The pharmacokinetics (ADMET) properties of strychnine were evaluated using SwissADME tool. Molecular docking was performed using Autodock Vina to explore the interactions between the active compounds and the target protein. RESULTS: Five strychnine toxicity-related components and a gene set of 40 genes were obtained. GO and KEGG analyses showed that Strychnine acts on the central nervous system through G protein-coupled receptor signaling pathway. Analysis of "ADMET" related parameters showed a high gastrointestinal tract absorption of (S)-stylopine and isobrucine and the compounds could cross the blood brain barrier. CHRM1 was selected as a key gene in strychnine toxicity. Molecular docking results showed that the co-crystalized ligands did not form hydrogen bond with CHRM1. (S)-stylopine had the highest binding affinity (binding energy = - 8.5 kcal/mol) compared with the other two compounds. CONCLUSION: Network toxicology and molecular docking reveal the toxicity mechanisms of strychnine active compounds. The findings showed that CHRM1 is a potential neurotoxic target. (S)-stylopine showed stronger neurotoxic effect compared with the other ligands.

RESUMO

Organic-inorganic halide perovskites are emerging materials for photovoltaic applications with certified power conversion efficiencies (PCEs) over 25%. Generally, the microstructures of the perovskite materials are critical to the performances of PCEs. However, the role of the nanometer-sized grain boundaries (GBs) that universally existing in polycrystalline perovskite films could be benign or detrimental to solar cell performance, still remains controversial. Thus, nanometer-resolved quantification of charge carrier distribution to elucidate the role of GBs is highly desirable. Here, we employ correlative infrared-spectroscopic nanoimaging by the scattering-type scanning near-field optical microscopy with 20 nm spatial resolution and Kelvin probe force microscopy to quantify the density of electrons accumulated at the GBs in perovskite polycrystalline thin films. It is found that the electron accumulations are enhanced at the GBs and the electron density is increased from 6 × 1019 cm-3 in the dark to 8 × 1019 cm-3 under 10 min illumination with 532 nm light. Our results reveal that the electron accumulations are enhanced at the GBs especially under light illumination, featuring downward band bending toward the GBs, which would assist in electron-hole separation and thus be benign to the solar cell performance. Correlative infrared-spectroscopic nanoimaging by the scattering-type scanning near-field optical microscopy and Kelvin probe force microscopy quantitatively reveal the accumulated electrons at GBs in perovskite polycrystalline thin films.

RESUMO

To explore the effects of urea dissociation on reducing false-positive results of  the Elecsys HIV combi PT assay. A retrospective analysis was used to evaluate the false-positive rate of the Elecsys HIV combi PT assay. Six false-positive sera, six positive sera and six sera from patients with early HIV infection were collected. Dissociation was performed using 1 mol/L, 2 mol/L, 4 mol/L, 6 mol/L, or 8 mol/L urea, and HIV screening assay were then detected to select the appropriate concentration of urea dissociation. Next, 55 false-positive sera and 15 sera from early HIV infection were used to verify the best concentration of urea to achieve dissociation. Retrospective analysis showed that the COI of the Elecsys HIV combi PT assay in false-positive sera ranged from 1.0 to 200.0, and approximately 97.01%(227/234) of false-positive sera were in the range of 1.0-15.0. The avidity index (AI) in positive and false-positive sera decreased as the urea dissociation concentration increased. When the dissociation concentration was 6 mol/L, the AI of false-positive serum was between 0.0234 and 0.2567, and the AI of early HIV infection sera was between 0.4325 and 0.5017. The difference in AI between false-positive and positive samples was significant. When negativity was defined as an AI of less than 0.3970, the sensitivity and specificity were 100.0% and 100.0%, respectively. Urea-mediated dissociation could significantly reduce the false-positive rate of the Elecsys HIV combi PT assay with a low COI. Our findings provided a reference for distinguishing positive and false-positive of the Elecsys HIV combi PT assay.

RESUMO

Aggressive adolescents are preferable in some Western cultures, whereas Confucianism places great emphasis on the inhibition of aggressive behaviors in Chinese culture. Using the longitudinal social network analysis, we used a sample of 1354 Chinese adolescents (54% boys, ages 12-15) who were followed over 1 year at three time points to examine the association between friendship dynamics and physical, verbal, and indirect aggression and the moderating role of gender. This study found the following: (1) Students who were verbally aggressive were regarded as attractive, whereas those who were indirectly aggressive were unattractive as friends; (2) adolescents selected peers with similar levels of aggression as friends; (3) adolescents were influenced by their friends' aggressive behaviors; and (4) girls were more susceptible than boys to the influence of physical aggression, although gender did not moderate the influence process of verbal and indirect aggression. The findings of this study provided a clearer insight into the selection and influence processes of the three subtypes of aggression and contributed to the diversity of samples. Chinese educators should pay more attention to both verbal aggression because of youths' preference for it and to girls with physically aggressive friends since they are more susceptible than boys.

RESUMO

Plasmonic optical antennas (POAs), often constructed from gold or silver nanostructures, can enhance the radiation efficiency of emitters coupled to POAs and are applied in surface-enhanced Raman spectroscopy (SERS) and light-emitting devices. Over the past four decades, radiation enhancement factors (REFs) of POA-emitter systems were considered to be difficult to calculate directly and have been predicted indirectly and approximately, assuming POAs are illuminated by electromagnetic plane waves without emitters. The validity of this approximation remains a significant open problem in SERS theory. Herein, we develop a method based on the rigorous optical reciprocity theorem for accurately calculating the REFs of emitters in nanoparticle-substrate nanogaps for single-molecule SERS and scanning probe-substrate nanogaps for tip-enhanced Raman spectroscopy. We show that the validity of the plane wave approximation breaks down if high-order plasmonic modes are excited. The as-developed method paves the way toward designing high-REF POA nanostructures for luminescence-related devices.

RESUMO

Plant phenolics are crucial defense phytochemicals against herbivores and glutathione S-transferase (GST) and carboxylesterase (CarE) in herbivorous insects are well-known detoxification enzymes for such xenobiotics. To understand relationship between a plant phenolic and herbivore GST or CarE genes, we evaluated the relationship between a rice phenolic ferulic acid and resistance to brown planthopper (BPH, Nilaparvata lugens), and investigated the interaction of ferulic acid with GST or CarE genes in BPH. The results indicate that ferulic acid content in tested rice varieties was highly associated with resistance to BPH. Bioassays using artificial diets show that the phenolic acid toxicity to BPH was dose dependent and the LC25 and LC50 were 5.81 and 23.30 µg/ml at 72 hr, respectively. Activities of the enzymes BPH GST and CarE were increased at concentrations below the LC50 of ferulic acid. Moreover, low ferulic acid concentrations (< LC25) upregulated the transcriptional levels of NlGSTD1 and NlGSTE1 of the GST family and NlCE of the CarE family. By using dsRNA-induced gene silencing (DIGS) of GST or CarE, it was shown that suppressed expression levels of NlGSTD1, NlGSTE1 and NlCE were 14.6%-21.2%, 27.8%-34.2%, and 10.5%-19.8%, respectively. Combination of NlGSTD1, NlGSTE1 or NlCE knockdown with ferulic acid increased nymph mortality by 92.9%, 119.9%, or 124.6%, respectively. These results suggest that depletion of detoxification genes in herbivorous insects by plant-mediated RNAi technology might be a new potential resource for improving rice resistance to BPH.

RESUMO

Lung cancer is a common disease with high mortality in China. Recent economic advances have led to improved medical capabilities, while costs associated with treating this disease have increased. Such change contributes to a commonly held belief that healthcare costs are out of control. However, few studies have examined this issue. Here, we use 34,678 hospitalization summary reports from 67 Guangxi hospitals (period 2013-2016) to document costs, temporal trends, and associated factors. Findings from this study are surprising in that they debunk the myth of uncontrolled healthcare costs. In addition, results and experiences from Guangxi are informative for other comparable regions.

RESUMO

BACKGROUND: The rising cost of healthcare is of great concern in China, as evidenced by the media features negative reports almost daily. However there are only a few studies from well-developed cities, like Beijing or Shanghai, and little is known about healthcare costs in rest of the country. In this study, we use hospitalization summary reports (HSRs) from admitted cardiovascular diseases patients in Guangxi hospitals during 2013-2016, and we investigate temporal trends of healthcare costs and associated factors. METHODS: By generalized additive model, we compute temporal trends of cost per stay (CPS), cost per day (CPD) and others. We then use generalized linear models to assess which factors associate with CPS and CPD. FINDINGS: Using a total of 760,000 HSRs, we find that CPS appears to be stabilized around $1040 until the middle of year 2015, before exhibiting a downward trend. Similarly, CPD exhibits similar stable pattern. Meanwhile, surgery-specific CPS showed an increase in year 2013-2014, and then stabilized. Drug costs account for over 1/3 of CPS, but they are gradually declining. Costs associated with physicians' and nurses' services represent less than 5% of CPS. We found that age, sex, marital status, occupation and payment methods are significantly associated with CPS or CPD. Interestingly, we found no association between patient ethnicity and these costs. However, we did find that minority patients use more secondary hospitals than Han patients. INTERPRETATIONS: Healthcare costs in Guangxi are stable, contrary to the rise portrayed by Chinese mass media. Several factors can be associated with healthcare costs, and these may be useful for developing evidence-based policies. In particular, there is a need to encourage more Han patients to seek care in primary and secondary hospitals.

RESUMO

Momordicin I and II are secondary metabolites from bitter melon (Momordica charantia L.) that are toxic to the Spodoptera litura ovary cell line (SL-1 cell). Both momordicin I and II significantly inhibited SL-1 cells proliferation. IC50 values after 24 h were 8.35 and 82.31 µg/mL, 6.11 and 77.49 µg/mL for 36 h, 4.93 and 49.42 µg/mL for 48 h for cells treated by momordicin I and II, respectively. IC50 values of the azadirachtin A control were 149.63, 54.54 and 23.66 µg/mL at 24, 36 and 48 h respectively, indicating that the cytotoxicity of momordicin I was significantly higher than that of momordicin II and azadirachtin A. Using inverted phase contrast microscopy we found that after 24 h exposure to momordicin I and II, cell shapes changed to circular, swelling increased, adherence ability declined and the cellular membrane bubbled. After 48 h exposure to momordicin I, most cells were suspended and dead; vacuole deformation and cytoplasm leakage indicated that momordicin I was more toxic to the cytoskeleton than momordicin II. Cells treated with momordicin I and II inhibited glucose absorption by 23.04 and 13.38% after 48 h and 47.60 and 20.92% after 60 h. Flow cytometry analysis suggested that SL-1 cells treated with momordicin I and II dramatically accumulated during the G2/M phase of the cell cycle, and total cell protein content increased by 56.93 and 35.81% respectively after 48 h treatment. Following treatment with momordicin I and II the karyotheca dissolved, the chromatin condensed abnormally and the nucleoli were damaged, migrated, or disappeared. The PI fluorescent value by FCM showed that the relative fluorescent intensity of SL-1 cells induced by momordicin I and II increased to 521.45 and 370.17, higher than 135.04 induced by control group treatment for 48 h. This indicated significant damage to the cytomembrane. Overall, the results demonstrate that suppression of cytoskeletal function, interference of mitotic figures and destruction of nuclear structure are effects of momordicin I and II exposure. These effects play major roles in momordicin I and II inhibition of SL-1 cells growth. The mode of action by which momordicins inhibit insect cell growth and development may be useful in the development of novel pest control formulations containing cucurbitane-type triterpene glycosides.

RESUMO

Phytochemical investigation of the ethanol extract from the leaves of Momordica charantia L. led to the isolation of two new (1, 2) and four known (3-6) cucurbitane-type triterpenoids. Their structures were elucidated on the basis of extensive analyses of spectroscopic data including IR, UV, MS, 1D, and 2D NMR. Also the absolute configurations of momordicines I (3) and II (4) were determined for the first time by application of the modified Mosher's method, acid hydrolysis, and GC analysis.

RESUMO

Southern rice black-streaked dwarf virus (SRBSDV), a non-enveloped icosahedral virus with a genome of 10 double-stranded RNA segments, is a novel species in the genus Fijivirus (family Reoviridae) first recognized in 2008. Rice plants infected with this virus exhibit symptoms similar to those caused by Rice black-streaked dwarf virus. Since 2009, the virus has rapidly spread and caused serious rice losses in East and Southeast Asia. Significant progress has been made in recent years in understanding this disease, especially about the functions of the viral genes, rice-virus-insect interactions, and epidemiology and control measures. The virus can be efficiently transmitted by the white-backed planthopper (WBPH, Sogatella furcifera) in a persistent circulative propagative manner but cannot be transmitted by the brown planthopper (Nilaparvata lugens) and small brown planthopper (Laodelphax striatellus). Rice, maize, Chinese sorghum (Coix lacryma-jobi) and other grass weeds can be infected via WBPH. However, only rice plays a major role in the virus infection cycle because of the vector's preference. In Southeast Asia, WBPH is a long-distance migratory rice pest. The disease cycle can be described as follows: SRBSDV and its WBPH vector overwinter in warm tropical or sub-tropical areas; viruliferous WBPH adults carry the virus from south to north via long-distance migration in early spring, transmit the virus to rice seedlings in the newly colonized areas, and lay eggs on the infected seedlings; the next generation of WBPHs propagate on infected seedlings, become viruliferous, disperse, and cause new disease outbreaks. Several molecular and serological methods have been developed to detect SRBSDV in plant tissues and individual insects. Control measures based on protection from WBPH, including seedbed coverage, chemical seed treatments, and chemical spraying of seedlings, have proven effective in China.

RESUMO

BACKGROUND: Southern rice black-streaked dwarf virus (SRBSDV) threatens rice production in China and Vietnam. The virus is vectored by the migrating white-backed planthopper (WBPH, Sogatella furcifera) in a circulative, propagative, and persistent manner. A persistently-transmitted plant virus might affect its vector's development and fecundity directly by infecting the vector itself and/or indirectly altering the host plant. This study evaluated the direct and indirect effects of SRBSDV on WBPH performance to better understand the virus-vector-host plant relationship in terms of its effects on the biological parameters of the vector. METHODS: Three experimental WBPH populations were established. Viruliferous and non-viruliferous populations were fed on SRBSDV-infected rice seedlings for 48 h as first-instar nymphs; infection status was confirmed by RT-PCR after they died. The control population was fed on healthy rice. Each insect was individually transferred to a healthy rice plant grown in a glass tube at 20°C, 25°C, or 28°C. Life parameters, including nymphal duration, survival rate, adult sex ratio, macropterous proportion, longevity, and oviposition amounts, of each population were measured at each temperature. RESULTS: The life parameter data indicated that SRBSDV and infected rice plants adversely influenced WBPH; the effects were temperature dependent. Compared with the control population, viruliferous populations showed significant changes, including prolonged nymphal stages and reduced survival rates at 20°C, while the non-viruliferous population had higher survival rates at 20°C and lower rates at 28°C compared with the control. Both populations had significantly shorter adult life spans at 25°C and lower oviposition amounts at 28°C relative to the control. CONCLUSIONS: Both SRBSDV-infection and feeding on infected rice plants affected vector performance. Although a longer nymphal period benefits viral acquisition and transmission by nymphs and might increase rice infection rate, in general, SRBSDV infection of the vectors and host plants was unfavorable to WBPH population expansion.

RESUMO

Plants have evolved complex processes to ward off attacks by insects. In parallel, insects have evolved mechanisms to thwart these plant defenses. To gain insight into mechanisms that mediate this arms race between plants and herbivorous insects, we investigated the interactions between gramine, a toxin synthesized by plants of the family Gramineae, and glutathione S transferase (GST), an enzyme found in insects that is known to detoxify xenobiotics. Here, we demonstrate that rice (Oryza sativa), a hydrophytic plant, also produces gramine and that rice resistance to brown planthoppers (Nilaparvata lugens, BPHs) is highly associated with in planta gramine content. We also show that gramine is a toxicant that causes BPH mortality in vivo and that knockdown of BPH GST gene nlgst1-1 results in increased sensitivity to diets containing gramine. These results suggest that the knockdown of key detoxification genes in sap-sucking insects may provide an avenue for increasing their sensitivity to natural plant-associated defense mechanisms.
